MC1R is one of those subjects where the details matter more than the headlines. This page pulls together the background, the mechanisms, and the practical points readers ask about most.
Updated 2026-02-14. Numbers and descriptions here follow the published literature rather than marketing material.
Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Reported observations after unregulated use include shifts in skin pigmentation and, in some accounts, unintended changes to moles and other lesions. Whether these outcomes are causally linked to the compound, and how often they occur, remain open questions because controlled data are scarce. The absence of standardised dosing and verified product purity complicates interpretation. Researchers have called for better surveillance and analytical characterisation of samples obtained outside regulated channels. Conclusions drawn from anecdotal evidence should be treated as provisional.
Melanotan-2 has not received marketing authorisation from major regulatory agencies for any therapeutic indication. Several jurisdictions classify it as a prescription-only medicine or a controlled substance when supplied for human use. Because approved products do not exist, material sold online usually sits outside pharmaceutical supply chains and formal quality oversight. Regulators have issued public notices describing the compound as unapproved. Enforcement varies, and the legal position differs between countries, which complicates any single general statement about its status.
Scientific discussion of Melanotan-2 spans pharmacology, dermatology, and public-health literature. Laboratory studies examine its receptor binding and cellular effects, while clinical reports describe outcomes observed after unregulated use. These two bodies of work differ in rigour and intent. Peer-reviewed trials of the compound as a medicine are limited, so much of the available information comes from case reports and surveillance data. Authors frequently note the gap between experimental findings and real-world use.

Melanocytes are the pigment-producing cells of the skin, and they carry melanocortin-1 receptors on their surface. When the receptor is activated, cyclic adenosine monophosphate rises inside the cell and raises the activity of enzymes such as tyrosinase, which increases melanin output. Melanotan-2 binds melanocortin-1 receptors in vitro and in animal models, and this binding is generally described as the basis for the tanning effect. Other receptors account for different effects: melanocortin-4 receptors contribute to appetite and erectile signalling, while melanocortin-3 and melanocortin-5 receptors contribute to energy balance and exocrine function.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Solid peptide material is generally stable when kept cold and dry. Common practice is storage at -20 degrees Celsius or lower, with desiccant and protection from light. Repeated freeze-thaw cycles and exposure to moisture are associated with degradation, aggregation, or loss of material. Once dissolved, stability depends on solvent, concentration, and temperature, and solutions are usually treated as short-lived unless stability data support longer periods. Handling notes typically emphasise minimising time at ambient temperature.
Identity and purity are assessed with chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the target peptide from related impurities and degradation products, and the resulting retention time is compared against a reference standard. Mass spectrometry, often coupled to liquid chromatography, confirms molecular mass. Amino acid analysis or peptide mapping can provide additional sequence-level confirmation when required. Results are only as reliable as the reference materials used alongside them.
=== Environmental === Both prenatal and perinatal obstetric complications may factor into the development of anorexia nervosa, such as preterm birth, maternal anemia, diabetes mellitus, preeclampsia, placental infarction, and neonatal heart abnormalities. Neonatal complications may also have an influence on harm avoidance, one of the personality traits associated with the development of AN. Neuroendocrine dysregulation: altered signaling of peptides that facilitate communication between the gut, brain and adipose tissue, such as ghrelin, leptin, neuropeptide Y and orexin, may contribute to the pathogenesis of anorexia nervosa by disrupting regulation of hunger and satiety. Gastrointestinal diseases: people with gastrointestinal disorders may be more at risk of developing disorders of eating practices than the general population, principally restrictive eating disturbances. An association of anorexia nervosa with celiac disease has been found. Individuals with good dietary management may develop anxiety, food aversion and eating disorders because of concerns around cross contamination of their foods. Some authors suggest that medical professionals should evaluate the presence of unrecognized celiac disease in all people with an eating disorder, especially if they present any gastrointestinal symptoms, (such as decreased appetite, abdominal pain, bloating, distension, vomiting, diarrhea or constipation), weight loss, or growth failure.
===== Bangladesh ===== American ties with Bangladesh were strained during Sheikh Hasina's rule, due to American accusations of human rights violations; India discouraged the United States from pressing too hard on these accusations to avoid giving ground to Islamist parties in future Bangladeshi elections. However, under Bangladesh's Yunus ministry, Bangladesh-US relations have improved, though Bangladesh's ties with India have worsened.

Corneal transplantation, also known as corneal grafting, is a surgical procedure where a damaged or diseased cornea is replaced by donated corneal tissue (the graft). When the entire cornea is replaced it is known as penetrating keratoplasty and when only part of the cornea is replaced it is known as lamellar keratoplasty. Keratoplasty simply means surgery to the cornea. The graft is taken from a recently deceased individual with no known diseases or other factors that may affect the chance of survival of the donated tissue or the health of the recipient. The cornea is the transparent front part of the eye that covers the iris, pupil and anterior chamber. The surgical procedure is performed by ophthalmologists, physicians who specialize in eyes, and is often done on an outpatient basis. Donors can be of any age, as is shown in the case of Janis Babson, who donated her eyes after dying at the age of 10. Corneal transplantation is performed when medicines, keratoconus conservative surgery and cross-linking can no longer heal the cornea. This surgical procedure usually treats corneal blindness, with success rates of at least 41% as of 2021.
=== Confirmation === In December 2015, the Joint Working Party of international scientific bodies International Union of Pure and Applied Chemistry (IUPAC) and International Union of Pure and Applied Physics (IUPAP) recognized the element's discovery and assigned the priority of the discovery to the Dubna–Livermore collaboration. This was on account of two 2009 and 2010 confirmations of the properties of the granddaughter of 294Og, 286Fl, at the Lawrence Berkeley National Laboratory, as well as the observation of another consistent decay chain of 294Og by the Dubna group in 2012. The goal of that experiment had been the synthesis of 294Ts via the reaction 249Bk(48Ca,3n), but the short half-life of 249Bk resulted in a significant quantity of the target having decayed to 249Cf, resulting in the synthesis of oganesson instead of tennessine. From 1 October 2015 to 6 April 2016, the Dubna team performed a similar experiment with 48Ca projectiles aimed at a mixed-isotope californium target containing 249Cf, 250Cf, and 251Cf, with the aim of producing the heavier oganesson isotopes 295Og and 296Og. Two beam energies at 252 MeV and 258 MeV were used. Only one atom was seen at the lower beam energy, whose decay chain fitted the previously known one of 294Og (terminating with spontaneous fission of 286Fl), and none were seen at the higher beam energy. The experiment was then halted, as the glue from the sector frames covered the target and blocked evaporation residues from escaping to the detectors.

Odum synthesized many of the concepts of ecosystem ecology, placing relationships between groups of organisms (especially material and energy relationships) at the center of the field. In the 1960s, as evolutionary theorists explored the possibility of multiple units of selection, ecologists turned to evolutionary approaches. In population ecology, debate over group selection was brief but vigorous; by 1970, most biologists agreed that natural selection was rarely effective above the level of individual organisms. The evolution of ecosystems, however, became a lasting research focus. Ecology expanded rapidly with the rise of the environmental movement; the International Biological Program attempted to apply the methods of big science (which had been so successful in the physical sciences) to ecosystem ecology and pressing environmental issues, while smaller-scale independent efforts such as island biogeography and the Hubbard Brook Experimental Forest helped redefine the scope of an increasingly diverse discipline.
=== Gene expression === PLP has been implicated in increasing or decreasing the expression of certain genes. Increased intracellular levels of the vitamin lead to a decrease in the transcription of glucocorticoids. Vitamin B6 deficiency leads to the increased gene expression of albumin mRNA. Also, PLP influences expression of glycoprotein IIb by interacting with various transcription factors; the result is inhibition of platelet aggregation.

=== Cavity-enhanced absorption spectrometry (CEAS) === The second way of improving the detectability of TDLAS technique is to extend the interaction length. This can be obtained by placing the species inside a cavity in which the light bounces back and forth many times, whereby the interaction length can be increased considerably. This has led to a group of techniques denoted as cavity enhanced AS (CEAS). The cavity can either be placed inside the laser, giving rise to intracavity AS, or outside, when it is referred to as an external cavity. Although the former technique can provide a high sensitivity, its practical applicability is limited because of all the non-linear processes involved. External cavities can either be of multi-pass type, i.e. Herriott or White cells, of non- resonant type (off-axis alignment), or of resonant type, most often working as a Fabry–Pérot (FP) etalon. Multi-pass cells, which typically can provide an enhanced interaction length of up to ~2 orders of magnitude, are nowaday common together with TDLAS. Resonant cavities can provide a much larger path length enhancement, in the order of the finesse of the cavity, F, which for a balanced cavity with high reflecting mirrors with reflectivities of ~99.99–99.999% can be ~ 104 to 105. It should be clear that if all this increase in interaction length can be utilized efficiently, this vouches for a significant increase in detectability.

Menkes disease is a congenital disease that is a cause of copper deficiency. Menkes disease is a hereditary condition caused by a defective gene involved with the metabolism of copper in the body. Menkes disease has a wide variety of symptoms including floppy muscle tone, seizures, abnormally low body temperature, and a peculiar steel color hair that feels very rough. Menkes disease is usually fatal, with the majority of affected children dying within the first ten years of life.
The ability of an ice road to safely support the weight of a vehicle, or any other loads applied onto it, referred to as bearing capacity, is the primary concern when designing, building and using that structure. Generally speaking, a vertically loaded ice cover will react in two ways: 1) it will sink, and 2) it will bend in flexure. In order to meet the ice bearing criteria, the top surface should not sink below the water line and the applied flexural stress should not exceed the ice's flexural strength. Three loading regimes have to be considered: a) maximum weight for standard usage or for parking during a short duration; b) a load that remains stationary during an extensive time period; and c) dynamic loading of the ice cover, from a traveling vehicle.
=== Neuronal proliferation === NGF can drive the expression of genes such as bcl-2 by binding to the Tropomyosin receptor kinase A, which stimulates the proliferation and survival of the target neuron. High affinity binding between proNGF, sortilin, and p75NTR can result in either survival or programmed cell death (PCD). Study results indicate that superior cervical ganglia neurons that express both p75NTR and TrkA die when treated with proNGF, while NGF treatment of these same neurons results in survival and axonal growth. Survival and PCD mechanisms are mediated through adaptor protein binding to the death domain of the p75NTR cytoplasmic tail. Survival occurs when recruited cytoplasmic adaptor proteins facilitate signal transduction through tumor necrosis factor receptor members such as TRAF6, which results in the release of nuclear factor κB (NF-κB) transcription activator. NF-κB regulates nuclear gene transcription to promote cell survival. Alternatively, programmed cell death occurs when TRAF6 and neurotrophin receptor interacting factor (NRIF) are both recruited to activate c-Jun N-terminal kinase (JNK); which phosphorylates c-Jun. The activated transcription factor c-Jun regulates nuclear transcription via AP-1 to increase pro-apoptotic gene transcription.

No regulator in a major market has approved it for human use. It appears in research settings and in products marketed outside pharmacy channels. Legal status for personal possession varies by country.
Afamelanotide is a linear analog that has received approval in some jurisdictions for a specific photoprotection indication. Melanotan II is cyclic and less selective across melanocortin receptors. The two are related but are not interchangeable in regulatory or clinical terms.
A compound that activates several receptor subtypes may produce effects beyond pigmentation. Selectivity influences which tissues respond and shapes the side-effect profile described in reports. Designing subtype-selective analogs is a stated goal of melanocortin research.
Major regulatory agencies have not approved it for any indication. Some countries permit it only under prescription frameworks, while others classify it as a controlled substance.
