Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.
Two structural changes distinguish the synthetic peptide from the natural hormone. A norleucine residue replaces methionine at one position, and a D-configured phenylalanine replaces the natural L-form at another. Both substitutions slow enzymatic breakdown, which extends the molecule's persistence relative to the parent hormone. The lactam bridge further constrains the backbone into a stable conformation. These features are standard design strategies in peptide chemistry and are not unique to this compound; they appear across many research peptides built for improved stability.
Analytical confirmation of identity relies on mass spectrometry, most often coupled to liquid chromatography. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and provides a purity estimate based on peak area. Electrospray ionization mass spectrometry then confirms the expected molecular mass, while tandem mass spectrometry can map the fragment sequence. For research-grade material, these two techniques together form the standard minimum. Purity figures reported by vendors are frequently not traceable to an independent laboratory.
Independent verification is central to quality control because the compound is not produced under pharmaceutical manufacturing standards. Third-party laboratories can measure purity, identity, residual solvents, and microbial contamination, though the scope of testing varies between services. Reported analyses of vendor samples have shown batch-to-batch variation in peptide content and the presence of truncated or oxidized species. How much of this variation reflects synthesis conditions versus storage and shipping is not well characterized. No harmonized reference standard exists for the material as sold.
Handling guidance for melanotan II follows general practice for small synthetic peptides rather than a product-specific monograph. Lyophilized powder is typically kept at minus twenty degrees Celsius or colder, protected from light and moisture, because warmth and humidity accelerate degradation. Once reconstituted, solutions are usually refrigerated and used within a short window, as hydrolysis and microbial growth both become concerns. Repeated freeze-thaw cycles are generally avoided. These conventions come from laboratory peptide chemistry and not from formal stability studies on this specific compound.

Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
Melanocytes are the pigment-producing cells of the skin, and they carry melanocortin-1 receptors on their surface. When the receptor is activated, cyclic adenosine monophosphate rises inside the cell and raises the activity of enzymes such as tyrosinase, which increases melanin output. Melanotan-2 binds melanocortin-1 receptors in vitro and in animal models, and this binding is generally described as the basis for the tanning effect. Other receptors account for different effects: melanocortin-4 receptors contribute to appetite and erectile signalling, while melanocortin-3 and melanocortin-5 receptors contribute to energy balance and exocrine function.
Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.
Identity testing for a cyclic peptide of this size usually relies on reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The mass spectrum confirms molecular weight, while the chromatographic trace indicates the proportion of related impurities. Tandem mass spectrometry can provide sequence-level information when fragmentation data are compared against a reference standard. Nuclear magnetic resonance is sometimes used to confirm the lactam bridge, although it requires more material and greater operator expertise than routine chromatographic methods.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Solid peptide kept dry at minus twenty degrees Celsius, shielded from light and moisture, is generally considered stable for extended periods. Solutions are divided into single-use aliquots and held at minus twenty or minus eighty degrees Celsius, because repeated freeze-thaw cycles promote aggregation and loss of material to container surfaces. Hydrolysis of the backbone and oxidation of tryptophan are the principal degradation routes in aqueous solution, and both accelerate at ambient temperature. Hygroscopic uptake after a vial is opened can also shift the actual mass weighed, which affects any concentration calculated from it.
==== Resignation of Juan Silva Villegas ==== On 24 February 2022, a motion of censure against the Minister of Transportation, Juan Silva Villegas, was signed by 33 parliamentarians and filed in the Congress of the Republic. This was created at the initiative of the Popular Force group, and also signed by other center-right or right-wing opposition groups such as Popular Renewal, Avanza País, Alliance for Progress and We Are Peru. The motion of censure refers to the appointment of the minister without him having "the necessary capacity and aptitude to perform the duties, nor having any type of experience, education, or training in public administration, much less in the area of transportation and communications." The parliamentarians emphasized that the Minister's management and performance had "an incalculable number of errors and illegalities." The desire to create a network and numerous acts of corruption are underscored by a report by the Comptroller General of the Republic, Nelson Shack, and the resigning Deputy Minister Fabiola Caballero. On 1 March 2022, a debate took place in Congress regarding the motion of censure. It began at 8 p.m. and was suspended two hours later around 10 p.m. with Pedro Castillo's announcement on Twitter of the resignation of his minister Juan Silva Villegas. On 5 March 2022, a few days after Juan Silva Villegas' resignation, President Castillo decided to appoint Minister of Transport and Communications to Nicolás Bustamante, who was sworn in that same night.
Alamethicin is a channel-forming peptide antibiotic, produced by the fungus Trichoderma viride. It belongs to peptaibol peptides which contain the non-proteinogenic amino acid residue Aib (2-aminoisobutyric acid). This residue strongly induces formation of alpha-helical structure. Occasionally a radiologic diagnosis of disc degeneration is made incidentally when a cervical X-ray, chest X-ray, or abdominal X-ray is taken for other reasons and the abnormalities of the vertebral column are recognized. The diagnosis of DDD is not a radiologic diagnosis, since the interpreting radiologist is not aware whether there are symptoms. Typical radiographic findings include disc space narrowing, displacement of vertebral bodies, fusion of adjacent vertebral bodies, and development of bone in adjacent soft tissue (osteophyte formation). An MRI is typically reserved for those with symptoms, signs, and X-ray findings suggesting a need for surgical intervention. Treatment may include physical therapy for pain relief, ROM (range of motion), and appropriate muscle/strength training, with emphasis on correcting abnormal posture, assisting the paravertebral (paraspinous) muscles in stabilizing the spine, and core muscle strengthening; stretching exercises; massage therapy; oral analgesia with non-steroidal anti-inflammatory agents (NSAIDS); and topical analgesia with lidocaine, ice, and heat. Immediate surgery may be indicated if the symptoms are severe or sudden in onset, or suddenly worsen. Elective surgery may be indicated after six months of conservative therapy with unsatisfactory relief of symptoms.
=== 20th century === Laudanum was used in home remedies and prescriptions, as well as a single medication. For example, a 1901 medical book published for home health use gave the following two "Simple Remedy Formulas" for "dysenterry" [sic]: (1) Thin boiled starch, 2 ounces; Laudanum, 20 drops; "Use as an injection [meaning as an enema] every six to twelve hours"; (2) Tincture rhubarb, 1 ounce; Laudanum 4 drachms; "Dose: One teaspoonful every three hours." In a section entitled "Professional Prescriptions" is a formula for "diarrhoea (acute)": Tincture opium, deodorized, 15 drops; Subnitrate of bismuth, 2 drachms; Simple syrup, 1⁄2 ounce; Chalk mixture, 11⁄2 ounces, "A teaspoonful every two or three hours to a child one year old." "Diarrhoea (chronic)": Aqueous extract of ergot, 20 grains; Extract of nux vomica, 5 grains; Extract of Opium, 10 grains, "Make 20 pills. Take one pill every three or four hours." The early 20th century brought increased regulation of all manner of narcotics, including laudanum, as the addictive properties of opium became more widely understood, and "patent medicines came under fire, largely because of their mysterious compositions". In the US, the Pure Food and Drug Act of 1906 required that certain specified drugs, including alcohol, cocaine, heroin, morphine, and cannabis, be accurately labeled with contents and dosage. Previously many drugs had been sold as patent medicines with secret ingredients or misleading labels.

In cancer, the genomes of affected cells are rearranged in complex or unpredictable ways. In addition to single-nucleotide polymorphism arrays identifying point mutations that cause cancer, oligonucleotide microarrays can be used to identify chromosomal gains and losses (called comparative genomic hybridization). These detection methods generate terabytes of data per experiment. The data is often found to contain considerable variability, or noise, and thus Hidden Markov model and change-point analysis methods are being developed to infer real copy number changes. Two important principles can be used to identify cancer by mutations in the exome. First, cancer is a disease of accumulated somatic mutations in genes. Second, cancer contains driver mutations which need to be distinguished from passengers. Further improvements in bioinformatics could allow for classifying types of cancer by analysis of cancer driven mutations in the genome. Furthermore, tracking of patients while the disease progresses may be possible in the future with the sequence of cancer samples. Another type of data that requires novel informatics development is the analysis of lesions found to be recurrent among many tumors.
=== Return to Prague === In November 1945, Smolková-Keulemansová returned to Prague to continue her studies. She completed grammar school and realized that the biggest struggle in her supplementary exams was chemistry, so she began to study chemistry at Charles University in Prague, leading to her lifelong devotion and love for the subject. She graduated from the Faculty of Natural Sciences at Charles University in 1952.
=== Phylogeny === The cladogram presented here illustrates the "family tree" of reptiles, and follows a simplified version of the relationships found by M.S. Lee, in 2013. All genetic studies have supported the hypothesis that turtles are diapsids; some have placed turtles within Archosauromorpha, though a few have recovered turtles as Lepidosauromorpha instead. The cladogram below used a combination of genetic (molecular) and fossil (morphological) data to obtain its results.
The formal evaluation of the quantitative evidence from two or more trials bearing on the same question. This most commonly involves the statistical combination of summary statistics from the various trials, but the term is sometimes also used to refer to the combination of the raw data. === Murder of Charmaine West === Rose is believed to have murdered Charmaine shortly before Fred's prison release date of 24 June 1971. She is known to have taken Charmaine, Anne Marie and Heather to visit Fred on 15 June, and it is believed that Charmaine was killed on or very shortly after this date. As well as forensic dentistry confirming that Charmaine had died while Fred was still incarcerated, further testimony from Tracey Giles' mother, Shirley, corroborated the fact that Charmaine had died before Fred's release. In her later testimony at Rose's trial, Shirley stated that her two daughters had been playmates of Charmaine and Anne Marie when her family lived in the upper flat of 25 Midland Road in 1971. Shirley further stated that after her family had vacated the flat in April 1971, she had brought Tracey to visit Charmaine on a day in June, only for Tracey to be told by Rose: "She's gone to live with her mother, and bloody good riddance!" Shirley Giles was adamant that Fred was still in prison on this occasion. As with the Giles family, Rose explained Charmaine's disappearance to others who enquired about her whereabouts by claiming that Fred's first wife, Catherine "Rena" West, had taken her eldest daughter to live with her in Bristol. She contradictorily informed staff at Charmaine's primary school that the child had moved with her mother to London. When Fred was released from prison on 24 June, he allayed Anne Marie's concerns for her sister's whereabouts by claiming Rena had collected Charmaine and returned to her native Scotland.

Cnidarians form a phylum of animals that are more complex than sponges, about as complex as ctenophores (comb jellies), and less complex than bilaterians, which include almost all other animals. Both cnidarians and ctenophores are more complex than sponges as they have: cells bound by inter-cell connections and carpet-like basement membranes; muscles; nervous systems; and some have sensory organs. Cnidarians are distinguished from all other animals by having cnidocytes that fire harpoon-like structures that are mainly used to capture prey. In some species, cnidocytes can also be used as anchors. Cnidarians are also distinguished by the fact that they have only one opening in their body for ingestion and excretion i.e. they do not have a separate mouth and anus. Like sponges and ctenophores, cnidarians have two main layers of cells that sandwich a middle layer of jelly-like material, which is called the mesoglea in cnidarians; more complex animals have three main cell layers and no intermediate jelly-like layer. Hence, cnidarians and ctenophores have traditionally been labelled diploblastic, along with sponges. However, both cnidarians and ctenophores have a type of muscle that, in more complex animals, arises from the middle cell layer. As a result, some recent text books classify ctenophores as triploblastic, and it has been suggested that cnidarians evolved from triploblastic ancestors.

== Overview == Thyrotropic cells (also called thyrotropes, or thyrotrophs) are endocrine cells in the anterior pituitary which produce thyroid-stimulating hormone (TSH) in response to thyrotropin-releasing hormone (TRH) from the hypothalamus. Thyroid-stimulating hormone, or thyrotropin, triggers the release of thyroxine (T4) and triiodothyronine (T3) from the thyroid gland. Thyrotropes comprise around 5% of the anterior pituitary lobe cells.
=== Chemical industry === Many chemicals exhibit a specific rotation as a unique property (an intensive property like refractive index or specific gravity) which can be used to distinguish it. Polarimeters can identify unknown samples based on this if other variables such as concentration and length of sample cell length are controlled or at least known. This is used in the chemical industry. By the same token, if the specific rotation of a sample is already known, then the concentration and/or purity of a solution containing it can be calculated. Most automatic polarimeters make this calculation automatically, given input on variables from the user.
Hyaluronic acid (; HA; conjugate base hyaluronate), also called hyaluronan, is an anionic, nonsulfated glycosaminoglycan distributed widely throughout connective, epithelial, and neural tissues. It is unique among glycosaminoglycans as it is non-sulfated, forms in the plasma membrane instead of the Golgi apparatus, and can be very large: human synovial HA averages about 7 MDa per molecule, or about 20,000 disaccharide monomers, while other sources mention 3–4 MDa. Medically, hyaluronic acid is used to treat osteoarthritis of the knee and dry eye, for wound repair, and as a cosmetic filler. These therapeutic uses are possible with modifications to HA, and more are still being developed. The average 70 kg (150 lb) person has roughly 15 grams of hyaluronan in the body, one third of which is turned over (i.e., degraded and synthesized) per day. As one of the chief components of the extracellular matrix, it contributes significantly to cell proliferation and migration, and is involved in the progression of many malignant tumors. Hyaluronic acid is also a component of the group A streptococcal extracellular capsule, and is believed to play a role in virulence. Chemical modifications to HA can improve certain properties such as biocompatibility, permeation, and sustained drug release. Common modification sites include the carboxyl, primary alcohol, and N-acetyl groups. These modifications include amidation, Ugi condensation, esterification, and etherification.

It is a synthetic seven-amino-acid peptide modeled on alpha-melanocyte-stimulating hormone. It carries two non-natural substitutions and a cyclic bridge that increase its stability relative to the natural hormone. It circulates as a research chemical and is not an approved medicine.
Melanotan-1, also called afamelanotide, is a shorter linear analog with a different amino acid sequence and no lactam ring. It has been evaluated in formal clinical programs, while melanotan-2 has not. The two are distinct molecules and are not interchangeable.
No. Bremelanotide is a related but distinct cyclic peptide that reached approved status for one specific clinical indication. Melanotan-2 is a separate molecule with its own sequence and properties. Shared ancestry in the melanocortin family does not make them the same substance.
The lyophilized powder is generally held at minus twenty degrees Celsius or below, away from light and moisture. Reconstituted solutions are typically refrigerated and used quickly. These practices derive from general peptide handling rather than a formal stability study.
