Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Outside regulated medicine, melanotan II circulates through online vendors as a research chemical, often marketed for tanning. Products sold this way vary widely in purity, concentration, and labeling accuracy, and independent testing has documented discrepancies. Published reports describe both pigment effects and adverse reactions, including nausea, flushing, and darkening of existing moles. Long-term safety data are sparse, and no large controlled trial has established a risk profile. Questions about cumulative effects on melanocytes remain unresolved in the literature.
Melanotan II is a synthetic peptide analog modeled on alpha-melanocyte-stimulating hormone, a naturally occurring signaling peptide involved in pigmentation. Its structure is a cyclic heptapeptide containing two non-natural substitutions, norleucine at position four and D-phenylalanine at position seven. These modifications resist enzymatic breakdown and extend the molecule's activity relative to the native hormone. The compound binds melanocortin receptors and is studied mainly as a pharmacological tool rather than a therapeutic product. It has never received approval as a medicine in any major jurisdiction.

Melanocytes are the pigment-producing cells of the skin, and they carry melanocortin-1 receptors on their surface. When the receptor is activated, cyclic adenosine monophosphate rises inside the cell and raises the activity of enzymes such as tyrosinase, which increases melanin output. Melanotan-2 binds melanocortin-1 receptors in vitro and in animal models, and this binding is generally described as the basis for the tanning effect. Other receptors account for different effects: melanocortin-4 receptors contribute to appetite and erectile signalling, while melanocortin-3 and melanocortin-5 receptors contribute to energy balance and exocrine function.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
Fatty acids are an integral part of the phospholipids that make up the bulk of the plasma membranes, or cell membranes, of cells. These phospholipids can be cleaved into diacylglycerol (DAG) and inositol trisphosphate (IP3) through hydrolysis of the phospholipid, phosphatidylinositol 4,5-bisphosphate (PIP2), by the cell membrane bound enzyme phospholipase C (PLC).
The city has 99 sports facilities including 46 gymnasiums, 23 sports fields, and 5 swimming pools. The Stade Océane (Ocean Stadium), inaugurated in July 2012, replaced the Stade Jules Deschaseaux. With 25,000 seats, it can host football matches as well as other sporting and cultural events. Basketball and Handball matches are playued in the Dock Océane hall (3600 seats) while ice hockey is played at the ice hockey rink (900 seats). Of the five swimming pools in the city, two are operated by the municipality: the CNH (which has an Olympic pool for competitions) and Les Bains Des Docks (which was designed by the architect Jean Nouvel). Le Havre has the largest free outdoor skatepark in France with approximately 7,000 m2 allocated to the urban Boardsport. The port infrastructure allows for many water activities such as sailing, fishing, canoeing, and rowing. Finally, the beach is a place for kitesurfing, windsurfing and surfing.
== History of method == The bacteria one-hybrid system has undergone numerous modifications since its inception in 2005. It ultimately arose as a variation of the bacteria two-hybrid system, conceived in 2000, which itself was inspired by the yeast one- and two-hybrid systems. Whereas the two-hybrid versions can assess both protein–protein interaction and protein–DNA interactions, the one-hybrid system specializes in the latter. Meng et al.’s B1H system differs from the two-hybrid version in two key respects. It uses a randomized prey library consisting of many (<2×108) unique potential target sequences and also adds a negative selection step in order to purge this library of self-activating clones. Although these ideas were borrowed from the original yeast one-hybrid system, they had not yet been applied to a bacterial host before 2005. As the technique grew in popularity, researchers amended their protocols to improve the B1H system. Designing the fusion construct (bait) to the omega, rather than the alpha, subunit of RNA polymerase has recently been favoured in order to improve the chimera’s stereochemistry and dynamic range. A zinc-finger domain on the fusion construct and its corresponding DNA target site, adjacent to the randomized prey sequence, has also been added to the increases affinity and specificity of protein–DNA interactions. This increased overall binding affinity allows for the characterization of even those DNA-binding domain proteins which interact weakly with a target sequence.
In analytical chemistry, a tandem mass tag (TMT) is a chemical label that facilitates sample multiplexing in mass spectrometry (MS)-based quantification and identification of biological macromolecules such as proteins, peptides and nucleic acids. TMT belongs to a family of reagents referred to as isobaric mass tags which are a set of molecules with the same mass, but yield reporter ions of differing mass after fragmentation. The relative ratio of the measured reporter ions represents the relative abundance of the tagged molecule, although ion suppression has a detrimental effect on accuracy. Despite these complications, TMT-based proteomics has been shown to afford higher precision than label-free quantification. In addition to aiding in protein quantification, TMT tags can also increase the detection sensitivity of certain highly hydrophilic analytes, such as phosphopeptides, in RPLC-MS analyses.
=== 23 April === Russia claimed to have captured the village of Ocheretyne, Donetsk Oblast, which was attributed to a rotational error by Ukrainian military commanders that left the sector mostly undefended and led to Russian forces advancing by five kilometers. The Ukrainian foreign ministry announced that it would suspend consular services to overseas Ukrainian men who were eligible for military service until 18 May, with the exception of those returning to Ukraine. The SBU arrested a resident of Kharkiv on suspicion of spying for Russia. UK Prime Minister Rishi Sunak announced a new military aid package for Ukraine that included 400 vehicles, 1,600 weapons and four million rounds of ammunition, along with £500 million in funding. The US Senate passed the bill authorising financial aid for Ukraine. The bill must now be signed into law by President Joe Biden.

== Advantages and disadvantages == The major advantage of CYTOF is the ability to investigate a larger number of parameters per panel than other cytometry methods. This allows a greater understanding of complex and heterogeneous cell populations, without the need for many complex and overlapping panels. Panels can include up to 45 antibodies, as opposed to the 10 that can be done in conventional flow cytometry but require great expertise to design. However, development of spectral flow cytometry has closed the gap between flow and mass cytometry in terms of the maximum number of antibodies that can be used. More antibodies per panel saves on time, allows understanding of a larger picture, and requires fewer numbers of cells per experiment, which is particularly advantageous when samples are limited such as with tumour studies. The use of the heavy metal isotopes also lowers background when compared to using fluorescent antibodies. Some cell types, such as myeloid cells, have high rates of autofluorescence that create a lot of background noise in flow cytometry. However, the rare heavy metal isotopes used are not present in biological systems, therefore there is very little or no background seen, and overall sensitivity is increased. The detection overlap between the different heavy metals is also very low compared to the overlap seen in fluorescent cytometry, which makes it much simpler to design a panel of many markers. Fluorescent dyes are subject to photobleaching, requiring the entire process to happen within a few hours after staining.
In June 2019, academic Stephen Wertheim called President Trump a "xenophobe" and criticised Trump's foreign policy toward China for heightening risks of a new Cold War, which Wertheim wrote "could plunge the United States back into gruesome proxy wars around the world and risk a still deadlier war among the great powers". In August 2019, Yuan Peng of the China Institute of International Studies said that the 2008 financial crisis "initiated a shift in the global order". Yuan predicted the possibility of the new cold war between both countries and their global power competition turning "from 'superpower vs. major power' to 'No. 1 vs. No. 2'". On the other hand, scholar Zhu Feng said that their "strategic competition" would not lead to the new Cold War. Zhu said that the US–China relations have progressed positively and remained "stable", despite disputes in the South China Sea and Taiwan Strait and US president Trump's aggressive approaches toward China. In January 2020, columnist and historian Niall Ferguson opined that China is one of the major players of this Cold War, whose powers are "economic rather than military", and that Russia's role is "quite small". Ferguson wrote: "[C]ompared with the 1950s, the roles have been reversed. China is now the giant, Russia the mean little sidekick. China under Xi remains strikingly faithful to the doctrine of Marx and Lenin.
The United States Armed Forces' Meal, Ready-to-Eat (MRE) is packaged similarly to the Canadian ration. Each sealed plastic bag contains one entire precooked meal, with a number of supplements and accessories. == History == The FDA's approval of sildenafil in 1998 was a ground-breaking commercial event for the treatment of ED, with sales exceeding US$1 billion. Subsequently, the FDA approved both vardenafil and tadalafil in 2003. It initially was developed by the biotechnology company ICOS, and then again developed and marketed worldwide by Lilly ICOS, LLC, the joint venture of ICOS Corporation and Eli Lilly and Company. Tadalafil was approved in 2009 in the United States for the treatment of pulmonary arterial hypertension and is under regulatory review in other regions for this condition. In late November 2008, Eli Lilly sold the exclusive rights to commercialize tadalafil for pulmonary arterial hypertension in the United States to United Therapeutics for an upfront payment of $150 million. Tadalafil was discovered by Glaxo Wellcome (now GlaxoSmithKline) under a partnership between Glaxo and ICOS to develop new drugs that began in August 1991. In 1993, the Bothell, Washington, biotechnology company ICOS Corporation began studying compound IC351, a phosphodiesterase type 5 (PDE5) enzyme inhibitor. In 1994, Pfizer scientists discovered that sildenafil, which also inhibits the PDE5 enzyme, caused penile erection in males participating in a clinical study of a heart medicine. Although ICOS scientists were not testing compound IC351 for treating ED, they recognized its potential usefulness for treating that disorder. Soon, in 1994, ICOS received a patent for compound IC351 (structurally unlike sildenafil and vardenafil), and Phase 1 clinical trials began in 1995.
== Treatment == Available treatment falls into two modalities: treating infections and boosting the immune system. Prevention of Pneumocystis pneumonia with trimethoprim/sulfamethoxazole is useful in those who are immunocompromised. In the early 1950s, immunoglobulin (Ig) was used by doctors to treat patients with primary immunodeficiency through intramuscular injection. Ig replacement therapy infusions can be either subcutaneous or intravenously administered, resulting in higher Ig levels for about three to four weeks, although this varies with each patient.
=== Extrapolating in pharmacology === In pharmacology, IVIVE can be used to approximate pharmacokinetics (PK) or pharmacodynamics (PD). Since the timing and intensity of effects on a given target depend on the concentration time course of candidate drug (parent molecule or metabolites) at that target site, in vivo tissue and organ sensitivities can be completely different or even inverse of those observed on cells cultured and exposed in vitro. That indicates that extrapolating effects observed in vitro needs a quantitative model of in vivo PK. Physiologically based PK (PBPK) models are generally accepted to be central to the extrapolations. In the case of early effects or those without intercellular communications, the same cellular exposure concentration is assumed to cause the same effects, both qualitatively and quantitatively, in vitro and in vivo. In these conditions, developing a simple PD model of the dose–response relationship observed in vitro, and transposing it without changes to predict in vivo effects is not enough.
The use of psilocybin mushrooms in religious ceremonies dating back thousands of years is contested. Despite popular narratives portraying psychedelics as ancient, widespread, and primarily used by shamans for therapeutic healing, anthropological and historical research shows their traditional use was limited, recent, and culturally specific, with modern Western interpretations largely shaped by idealization, tourism, and ideological agendas. Reliable evidence shows that psilocybin mushrooms were used ritualistically in pre-Columbian Mexico but were otherwise rare, with most claims of ancient widespread use exaggerated or misinterpreted. It has been argued that the Tassili Mushroom Figure, discovered in Tassili, Algeria, is evidence of an early psilocybin-containing mushroom cult. 6,000-year-old pictographs discovered near Villar del Humo, Spain, illustrate several mushrooms that have been argued to be Psilocybe hispanica, a hallucinogenic species native to the area. Some scholars have also interpreted archaeological artifacts from Mexico and the so-called Mayan "mushroom stones" of Guatemala as evidence of ritual and ceremonial use of psychoactive mushrooms in the Mayan and Aztec cultures. After Spanish conquistadors of the New World arrived in the 16th century, chroniclers reported mushroom use by the natives for ceremonial and religious purposes. According to the Dominican friar Diego Durán in The History of the Indies of New Spain (published c.
Ben Gibbons of Screen Rant criticized shortening the episodes comparing to the previous seans, arguing that the first three episodes have been "jumping from one narrative to another, trying to create loose connections between them all, but struggling to make one consistent thread that captivates the audience". Writing for Slant Magazine, Michael Savio called the season a "nostalgia-ridden nightmare as mildly engrossing as it is exceedingly gross". He felt that Lange was "mesmerizing any time she's on screen", but when she "isn't holding our attention, these early episodes are borderline incoherent, if not just plain boring".

No. It is a synthetic analogue carrying four amino acid changes, a lactam ring and an amidated C-terminus. The natural hormone is a linear thirteen-amino-acid peptide processed from proopiomelanocortin.
Afamelanotide is the linear analogue [Nle4-D-Phe7]-alpha-MSH, sometimes called melanotan I, while melanotan II is cyclic and carries three further substitutions. The two are distinct molecules and are not interchangeable in analytical testing.
Binding to the four melanocortin receptor subtypes is well documented in vitro. The relative contribution of each subtype to whole-body effects in humans is far less certain.
It is a synthetic cyclic peptide designed as an analog of alpha-melanocyte-stimulating hormone. It acts on melanocortin receptors and is best known from research into pigmentation. It is not an approved pharmaceutical product.
