This is a working overview of cyclic peptide, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2025-11-18. Anything still debated is marked as such rather than presented as settled.
Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.

Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
== Function == EBP functions as a Δ8–Δ7 sterol isomerase, catalyzing the migration of the double bond in the sterol B-ring from the 8(9) to the 7(8) position. In the Bloch pathway of cholesterol biosynthesis, EBP converts zymosterol to dehydrolathosterol, while in the Kandutsch–Russell pathway it converts zymostenol to lathosterol.
== Introduction == Banting lived at the house in London for ten months, beginning in June, 1920. He attempted a private medical practice and when it was unsuccessful, he began working at the University of Western Ontario; his research for a lecture there was what inspired his 25 word idea that provided the key to discovering insulin, and prevented the certain death of those affected by diabetes. Banting returned to the University of Toronto to begin his research on insulin in the spring of 1921. Banting House is dedicated to the story behind the discovery of insulin, as well as Banting's life and career. Its galleries focus on everything from his time spent in London, to his contributions in both World Wars, to his efforts as an artist. Some notable artifacts include Banting's original art, desk, medicine cabinet, and bed frame, as well as his Military Cross, the KBE, and his official replica of the Nobel Prize. Banting House was designated a National Historic Site of Canada in 1997.
Cheese curds, drained of the whey and served without further processing or aging, are popular in some French-speaking regions of Canada, such as Quebec, parts of Ontario, and Atlantic Canada. These are often sold in snack-sized packaging and seen as a typically Canadian food item. Throughout Canada cheese curds are served with french fries and gravy in a popular snack called poutine. Curds are also typical of some Germanic-descent regions such as historic Waterloo County in Ontario. In some parts of the Midwestern U.S., especially in Wisconsin, curds are eaten fresh without further additions, or they are breaded and fried.
α2-Macroglobulin is known to bind zinc, as well as copper in plasma, even more strongly than albumin, and such it is also known as transcuprein. 10 to 15% of copper in human plasma is chelated by α2-macroglobulin.
EMA has a lower sensitivity, but its specificity is near 100%; it can be used to confirm coeliac disease in those who have borderline TG2 IgA levels. EMA testing is costly, hard to interpret, and vulnerable to inter-observer and inter-site variability. DGP IgG is used to evaluate coeliac disease in those with IgA deficiency. Coeliac disease is more common in those with IgA deficiency, so medical guidelines recommend that people being tested for coeliac disease are also tested for IgA deficiency. Because IgA-based tests are unreliable in those with IgA deficiency, IgG-based tests are used instead. These include EMA IgG, DGP IgG, and TTG IgA, which are less accurate than IgA testing. Multiparametric serological assays allowing simultaneous detection of TG2 IgA and total IgA have been proposed to improve screening efficiency for coeliac disease. A study evaluating the Polycheck ® Celiac IgA + total IgA test reported high sensitivity and specificity for TG2 IgA and total IgA measurements in coeliac disease diagnostics. A 2020 guideline by the European Society of Paediatric Gastroenterology, Hepatology, and Nutrition (ESPGHAN) suggests biopsy can be avoided in children who have symptoms of coeliac disease, TTG IgA levels ten times higher than normal, and a positive EMA antibody. There is insufficient evidence to suggest that a nonbiopsy approach can be used in adults. Genetic testing is not needed to diagnose coeliac disease, but is sometimes used to clarify discrepancies between blood tests and histology.

PGC1-α (PPARGC1A), a transcriptional coactivator of nuclear receptors important to the regulation of a number of mitochondrial genes involved in oxidative metabolism, directly interacts with MEF2 to synergistically activate selective slow twitch (ST) muscle genes and also serves as a target for calcineurin signaling. A peroxisome proliferator-activated receptor δ (PPARδ)-mediated transcriptional pathway is involved in the regulation of the skeletal muscle fiber phenotype. Mice that harbor an activated form of PPARδ display an "endurance" phenotype, with a coordinated increase in oxidative enzymes and mitochondrial biogenesis and an increased proportion of ST fibers. Thus—through functional genomics—calcineurin, calmodulin-dependent kinase, PGC-1α, and activated PPARδ form the basis of a signaling network that controls skeletal muscle fiber-type transformation and metabolic profiles that protect against insulin resistance and obesity. The transition from aerobic to anaerobic metabolism during intense work requires that several systems are rapidly activated to ensure a constant supply of ATP for the working muscles. These include a switch from fat-based to carbohydrate-based fuels, a redistribution of blood flow from nonworking to exercising muscles, and the removal of several of the by-products of anaerobic metabolism, such as carbon dioxide and lactic acid. Some of these responses are governed by transcriptional control of the fast twitch (FT) glycolytic phenotype.
The French Angel (Maurice Tillet, 1903–1954), Russian-born French professional wrestler, is better known by his ring name, the French Angel. Pío Pico, the last Mexican Governor of California (1801–1894), manifested acromegaly without gigantism between at least 1847 and 1858. Some time after 1858, signs of the growth hormone-producing tumor disappeared along with all the secondary effects the tumor had caused in him. He looked normal in his 90s. His remarkable recovery is likely an example of spontaneous selective pituitary tumor apoplexy. (Leonel) Edmundo Rivero, Argentine tango singer, composer and impresario. The Unionists, by and large, regarded Germany as a dangerous rival, and rejoiced at the chance to destroy her. They meant to fight a hard-headed war by ruthless methods; they condemned Liberal 'softness' before the war and now. The Liberals insisted on remaining high-minded. Many of them had come to support the war only when the Germans invaded Belgium. [...] Entering the war for idealistic motives, the Liberals wished to fight it by noble means and found it harder to abandon their principles than to endure defeat in the field. The 1915 coalition fell apart at the end of 1916, when the Conservatives withdrew their support from Asquith and gave it instead to Lloyd George, who became prime minister at the head of a new coalition largely made up of Conservatives. Asquith and his followers moved to the opposition benches in Parliament and the Liberal Party was deeply split once again.
== Philosophical background == Traditional Chinese medicine includes a broad range of practices sharing common concepts that developed in China for more than 2,000 years, including various forms of herbal medicine, acupuncture, massage (tui na), exercise (qigong), and dietary therapy. It is primarily used as a complementary alternative medicine approach. Traditional practices are widely used in China and, in an adapted form, also in the West. Its philosophy is based on School of Yin Yang, which incorporates Five Phases theory and Yin–Yang theory, and elements of which were absorbed by Daoism. Philosophical texts influenced medical thought and practice, mostly by being grounded in the same theories of qi, yin-yang and wuxing and microcosm-macrocosm analogies.

== Use in stem cell biology == Thy-1 can be considered as a surrogate marker for various kind of stem cells (e.g. hematopoietic stem cells or HSCs). It is one of the popular combinatorial surface markers for FACS for stem cells in combination with other markers like CD34. In humans, Thy-1 is expressed on neurons and HSCs among others. It is considered a major marker of HSC pluripotency in concordance with CD34. In human HSCs, Thy1 cells are all CD34 positive. Thy 1 is also a marker of other kind of stem cells, for example: mesenchymal stem cells, hepatic stem cells ("oval cells"), keratinocyte stem cells, putative endometrial progenitor/(?)stem cells.
== Side effects == The side effects of flutamide are sex-dependent. In men, a variety of side effects related to androgen deprivation may occur, the most common being gynecomastia and breast tenderness. Others include hot flashes, decreased muscle mass, decreased bone mass and an associated increased risk of fractures, depression, and sexual dysfunction including reduced libido and erectile dysfunction. In women, flutamide is, generally, relatively well tolerated, and does not interfere with ovulation. The only common side effect of flutamide in women is dry skin (75%), which can be attributed to a reduction of androgen-mediated sebum production. General side effects that may occur in either sex include dizziness, lack of appetite, gastrointestinal side effects such as nausea, vomiting, and diarrhea, a greenish-bluish discoloration of the urine, and hepatic changes. Because flutamide is a pure antiandrogen, unlike steroidal antiandrogens like cyproterone acetate and megestrol acetate (which additionally possess progestogenic activity), it does not appear to have a risk of cardiovascular side effects (e.g., thromboembolism) or fluid retention.
Creation of the benzoquinone structure (using phenylalanine or tyrosine, via 4-hydroxybenzoate) Creation of the isoprene side chain (using acetyl-CoA) The joining or condensation of the above two structures The initial two reactions occur in mitochondria, the endoplasmic reticulum, and peroxisomes, indicating multiple sites of synthesis in animal cells. An important enzyme in this pathway is HMG-CoA reductase, usually a target for intervention in cardiovascular complications. The "statin" family of cholesterol-reducing medications inhibits HMG-CoA reductase. One possible side effect of statins is decreased production of CoQ10, which may be connected to the development of myopathy and rhabdomyolysis. However, the role statins play in CoQ deficiency is controversial. Although statins reduce blood levels of CoQ, studies on the effects of muscle levels of CoQ are yet to come. Genes involved include PDSS1, PDSS2, COQ2, and COQ8A. Organisms other than humans produce the benzoquinone and isoprene structures from somewhat different source chemicals. For example, the bacteria E. coli produces the former from chorismate and the latter from a non-mevalonate source. The common yeast S. cerevisiae, however, derives the former from either chorismate or tyrosine and the latter from mevalonate. Most organisms share the common 4-hydroxybenzoate intermediate, yet again uses different steps to arrive at the "Q" structure.
In the 2011–12 season, a coaching change by appointing Christian Streich, with the club finishing 12th. Under Christian Streich, the 2012–13 Bundesliga season saw the club finish in fifth place, their best league standing since 1994–95. The fifth-place finish secured a position in the 2013–14 UEFA Europa League. Had Freiburg defeated Schalke 04 on the final matchday of the season, Freiburg would have advanced further in the league table against Schalke and qualified for the UEFA Champions League for the first time in club history. The 1–2 defeat to Schalke, however, saw Schalke secure fourth place in the league and qualify for the tournament instead. During the 2012–13 season, Freiburg also advanced to the semi-finals of the DFB-Pokal for the first time in the club's history, but lost to local rivals VfB Stuttgart 1–2, and missed the chance to play Bayern Munich in the final. In the 2014–15 season, after six years in the top flight, Freiburg was relegated to the 2. Bundesliga by a single point after a final-day defeat at Hannover 96. This was despite beating Bayern Munich in the second-last game. In the following season, however, the club earned its fifth promotion to the Bundesliga, with two matches to spare. The first season back in the Bundesliga saw them end seventh. This saw Freiburg qualify for the Europa League, as German cupwinners Borussia Dortmund were already qualified for the Champions League. The side were eliminated in the third qualification round against NK Domžale from Slovenia. Freiburg stayed in the top flight, finishing 15th.

It is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone, containing seven amino acids with a lactam ring and a D-configured phenylalanine residue. It is supplied as a lyophilised powder for laboratory research.
The natural hormone is a linear tridecapeptide that is rapidly degraded in circulation. Melanotan-2 is shortened, cyclised, N-terminally acetylated, and carries a D-amino acid substitution, all of which slow enzymatic breakdown.
No national medicines regulator has approved melanotan-2 for therapeutic or cosmetic use. In several jurisdictions it is treated as an unapproved prescription medicine, and its legal classification differs from country to country.
It is a synthetic cyclic heptapeptide and an analogue of alpha-melanocyte-stimulating hormone. The molecule is produced by chemical synthesis rather than extracted from a biological source.
